Short answer-
It's definitely possible using Session-at-Once recording, but it might not be necessary to burn more than one CD in the first place, depending on what your needs are.
More information-
There's a number of different ways to go about this, but you haven't provided even close to enough info. How big is the source file, which version of Nero are you using, and what do you plan on playing this back on? Also, what OS are you using and what version of Windows Media Player are you using? This is important due to Digital Rights Management.
If you're planning on playing this back on an older CD player, you might very well have to burn multiple CDs. However, if your car stereo has the ability to play MP3s (most newer ones do), you could convert the source file to MP3 if it isn't already, and burn it as a single data disc. You'd most likely have plenty of space left over on the disc for either more audio or other data files you'd like to back up as well.
People have reported lots of playback problems with CDs they've burned after upgrading to either Windows Vista or Windows Media Player 11 where there weren't any before. This is especially true when their CD player is unable to play protected files, such as those purchased from an online store.
