Yeah looks like we can already figure out the whole plot.
Potentially predictable and disappointing story aside, it's nice to see that AC has recovered from the franchise fatigue that affected Brotherhood and Revelations.
I guess I won't be skipping AC3 after all.

I really need to pick up Brotherhood and Revelations to get caught up all the story. Or could I just skip those and play III and be fine with the main story?

No way. I recommend AC I and II, wiki/YouTube brotherhood because it sucked and play Revelations.

Brotherhood was a step above 2, Revelations was a side step from Brotherhood; both better and worse in various areas. Really, you could just hit up the wiki to get ready for 3. The amount of storyline relevant to understanding 3 isn't more than a few paragraphs for each game.

What this guy said. However, the underground dungeons in Revelations (which are basically a test of your knowledge of the game) had some of the most badass real-time events I've ever seen in a game.

If by main story you mean Desmond's story, just watch the Youtube clips of the endings. They both had minor details during the game, but like 1 and 2, the ending is where I'd say 90%+ of the plot development in Desmond's story occurred.

By the way, there's been mention of 2-4 player co-op... I'm assuming that's some kind of mission mode, but has anything been specifically explained? I would love to be able to go through the story online with my friend seeing as I'm moving next month D: We've been playing these games together sittin on his couch for years..
