Just got the offer to join the Brotherhood of Steel. He made it sound like I'd be giving up other options, so I decided to hold off on the decision for now.

Do I lock out other questlines by choosing the Brotherhood route? I'm not done with either the Railroad or the Minutemen quest lines yet (but I've started both), if that makes a difference. Or could I potentially finish those quest lines and come back and do Brotherhood?

I'm sure I could figure it out with some research, but I'm a little wary of spoilers.

You dont lock anything until near the end of the game when you have to choose a faction that usually wants to kill the other factions.
If you are far enough in the story to unlock the railroad then you are fairly far in the game.

You can run missions with all of them. But like Tek said the factions will ask you at one point to kill the other faction.

One of the Institute's quests makes you pick the Institute or the Brotherhood. The other remains hostile to you the rest of the game.

It will even say "doing this will make faction x hostile. Do you want to proceed? Y/n"

Unless something changed that option doesn't come up for the Brotherhood version. You'll talk to an NPC and in the middle of the conversation the message will just pop up that the railroad is hostile.

Follow the main storyline. You can get into the institute with the help of BoS.

There's multiple ways to the Institute, just depends on which faction (can also choose no faction) you end up siding with.
