There's a valid question of whether Serena has dominated women's tennis more thoroughly than any man has dominated men's, just like you can argue that Mayweather dominated his weight class more than Muhammad Ali dominated his - but the raw question of "Is Serena the best tennis player" without qualification returns the answer of "of course not" in the same way "Is Mayweather the best boxer" without qualification does the same.

SPORTS TALK Ali faced a higher level of competition but it's not arguable, Mayweather was unquestionably more dominant at his natural weight classes than Ali was at heavyweight. when discussing cross-gender and cross-weight talk of best though it's understood the concept is in a pound for pound sense, that is an athlete's physical and mental skills irrespective of their size. in an absolute sense the biggest great fighters will usually be the absolute best, as you say.

some wouldn't qualify it (with women's) - some would just say she's the best in the world

i mean Serena is arguably the most physically imposing woman in the history of women's tennis and she's 5'9" and 155 lbs which makes her a little small for a high school men's varsity team, none the less the fully grown space marines that are modern male professional athletes. after a certain point of even remotely comparable skill the size and strength gap is just way too large to overcome.
