Basically each casino has odds makers on staff that analyze every game about 2-3 weeks out and come up with the lines. They tinker with them based on the teams' results and injuries after the initial internal line is made, and then they go up on the board as soon as the teams involved don't have any more games left. That's the "open" number.

After that, based on other casinos' opening lines, number of bets placed on a certain side, and other factors, the line will move mid-week. It's usually only a point or two at most unless someone gets hurt/suspended/etc unexpectedly.

Individual games don't usually move much based on how many people bet a certain side. For every 30,000 people throwing down $100 on the Boys, there's 3 guys dropping $1mil on Carolina. Big shifts in a line without an obvious reason means both the big bettors and the public agree, which can be a big red flag. Either Vegas knows something no one else does or their oddsmakers are just off, and it's usually the former. The top players with have $5-10 mil live on the Super Bowl or other big games, and will place big bets they think will lose in order to move the line into a favorable position to go really big with an extra few points.

However futures bets, like picking Super Bowl winners in August, are very influenced by how popular teams are. Sexy picks like Cowboys, Cubs, Notre Dame, etc are almost never good futures bets.

There was a 60 Minutes a few years ago about one of the big players in Vegas, I'm out of town but I can dig it up if you actually want to see it.
