While there's been some great advice in this topic, I'm going to throw in my two cents. I've had really good success with girls, so hopefully I can pass along my wisdom.
The call time is not as important as people put pressure on it. I've had great success waiting a few days to call a girl. Once, when I was waiting tables, a girl left me her number, and I called her the moment she walked out the door, on my cell. Both instances worked with great success. The only thing that really can give you determination about this sort of thing is based on how you act when you call her. Remember to make her feel appreciated for giving you her number - why girls have anything to do with us stupid boys, I'll never know. But also keep in mind the importance of seeming self-confidant, even if you're about to pee your pants from nervousness - I've been in those instances too.
The first key to winning with a girl is to never let on how nervous you may be. One of the things which girls appreciate at first is deception. I'm not talking about outright lying, but more so, intrigue. Don't rush into talking about yourself much, let her do most of the talking. Consider conversation to be like fishing. You throw out the lure of topic, and let her nibble and bite. I hate to relate dating girls to such a terribly boring sport, but it's quite true. Most girls I've dated were always puzzled by the fact that I didn't let on a whole lot about myself. Hell, even my girlfriend who I've been with for over a year now doesn't know a whole lot about me - and we live together!
As I've seen it stated elsewhere, remember, when you're dealing with girls, the number one rule is: It's all about her. When you ask her out, most likely she'll ask you what you want to do. When you offer the option, for example, of going to the movies, don't just say, "Well, we could go to the movies if you like." Instead try, "Well, are you into movies?" (pause and wait for a "Yes.") "Well, there's a couple things out right now that I think look pretty good. There's 'X movie' with 'x actor' that came out last weekend. I've been hearing good things. Then again, 'Y movie' looked pretty good in the previews." And that way you've controlled what you do, by allowing her to feel as if she's making all the choices.
Most girls love having power - but don't like taking control. Make sense?
Anyway, these little tips should get you to your first date. I don't know how much experience you have there, but ultimately, just be yourself. Unless you're overly nervous. I can't begin to stress the importance of the word "suave." Any way you slice it, either the girl will appreciate you for who you are - which seems to be the case thus far - or, she'll never see you again. In either instance, don't forget there's a shit ton of girls in this world. Hot, gamer girls are aplenty nowadays.
And if you get ditched, you could always turn to your friend 4chan. Or slip her a mickey when she's not looking. In that option, see wisdom above for pleasures below.

You don't have to follow the three-day rule. I met my current girlfriend by calling her two days later, and we've been together since the summer.
As far as what to talk to her about, start off with the general "how was your day?" "what are you up to?" "what have you been up to?" and when she tells you what she has been up to, you build off of that. If she tells you that she went to class, ask her about what she's studying for. If she tells you she was at work, ask her where she works and how she likes the jobs and just build off of that.
Act natural, it's really not as hard as people make it out to be.
At least from the chick perspective.. don't wait three days. If there was a total stranger that I hit it off with enough to say "Hey, I'm actually going to be proactive and OFFER this gentleman a way to contact me", then I'm clearly interested in the interaction.
The fact that she gave YOU her number says pretty clearly both in ovary-speak and human means that "I would like to talk!". I would wait 'til the next afternoon, and then call her. Definitely do what Wafik said and THANK HER for the previous conversation, and make it clear that you're just as interested in a continuation.
Since she went out on a limb and made the overture to you, make sure you call her with a number of suggestions in mind for meeting up. Coffee is a fantastic start.
[edit 'cause I thought of something else]: Suggesting a movie is okay, but its just that.. okay. I'm always a little turned off by the idea of staring at a screen in a dark room for two hours as a means of "hanging out" with someone. Sure, you have a bunch of stuff to talk about afterwards, but you just paid $18 to NOT talk to them for the length of the movie.
Three days is way too long. But then again the last time I gave my number out I made the mistake of doing so before the end of the evening. By the time I left he had literally asked me 10 times when he got to drive me home, and the last time I said no and left, I had a message from him waiting when I got there. One of my friends from work thought it was cute... I was completely freaked out. The funnest, sometimes scariest part of dating is there are no real rules. Be yourself and see if you click. movies are terrrible first dates. You don't get a feel for the person and it will most likely cost you money (you should pay for her if its first date). Dinner also isnt the *best* idea since it sort of keeps you locked down in one area for a while so alot could go wrong. The best first dates are walking ones, like in a mall or along a street or something since you can talk about anything and there is plenty of subject material.
