While it's good that you asked, it does sound like you don't have much chance in the audition. It sounds like your director feels that "looking the part" is particularly important for the role. So goes show business.

Here's what you do. Absolutely smash the audition so that the director asks you to take the lead. Then politely decline and tell him you're happy with the role you are in and just wanted to prove to yourself that you could hold a lead role.
That'll not only teach him to pick the right person the first time, but it will show ambition.
Note: Don't really do that.
My sister is in London doing her degree in Dance and Drama (I think...) and she pretty much has to do what all these guys are saying. Just go for it. Ignore what hes saying about you not being imagined for the role, just belt your heart out and let fate do the rest. Theres been many a time she hasn't been shortlisted for a part and people worse than her have, then she'll get a callback later on in the week, surprise them and get the part. Its sooo up and down in your profession, just gotta roll with it.
If you don't get it, it just wasn't meant to be, there'll be other parts etc etc

Just an update: the mini-audition is tomorrow, the producer sent me an email for it filled with exclamation points and such so I hope that's a good sign.
The director emailed me back with this today after some discussion:
"No matter the outcome of tomorrow’s mini-audition, I want you to know that you are being STRONGLY considered for this role now and that “body type” and “age” do NOT matter. Come and have fun, no matter what we decide in the end."
I am about 10 yrs older than the part "should" be, and I am not typical leading lady type (ie, my boobs are too big and I'm too tall), so those were some concerns I expressed.
Tonight, dying my hair blonde to be more type and going to a cast party. Where I can't drink because tomorrow I have to go sing at noon. LOL
The sad thing is I WILL REALLY NEED THE DRINK! lol
Thanks for the advice guys, and all the well-wishes and helping me get through my neuroticism, I always go a bit manic when I'm not sure what to do and overanalyze everything. So I do appreciate everyone's respect and whatnot.

You have a great voice so I don't see how you can't get the part if they're saying that age and body type don't matter. At this point I think the only thing that can stop you from getting the part is being too nervous and messing up your audition (fumbling words for example).
Don't over analyze so much and go for it. Fear is good, it can motivate you, but fear of failure is very unhealthy. It's better to just give it your all and show what you can do. Worse case you won't get the part, but you won't be stuck saying "maybe I should have tried harder" and kicking yourself for not trying. Sometimes you gotta jump in the pool without testing the water.
