Do they say that during the Wrath explanation? The comic you showed said they would complete a human homunculus, not that it was a relatively new idea (So not definitive).
It's not that important really, though sort of interesting.

They do during King Bradley experimentation. They made a huge deal that Bradley was differents in both series, so unless you want to imagine an explanation that isn't needed, it's pretty clear to me that Bradley was the first, and that Greed was the 2nd attempt (althought the body didn't merge with the philosopher stone this time)

he's the only homunculus that can't regenerate either, which doesn't seem to have anything to do with him being human based.

Hohenheim is basically a human based homunculus, and he doesn't age. Probably because he has a shit load of souls to regenerate himself with, where as Bradley does not.

I thought The Little One in the Flask broke down his body and rebuilt him a new immortal one so that he could take information from his body to make one for himself?

What chapter had Bradley's backstory? I don't remember anything specific on him being the first human based homunculus.

I'm not sure if the anime said that, but the manga definitely didn't. All it says is that Father used information from Hohemheims blood to create his own body/container. And Father was created using Hohenheim's blood in the first place, so he shouldn't have needed to take it from him during the Xerxes transmutation event.

As far as I can tell, Hohenheim absorbed the souls/philosopher stone just from being at the center of the transformation circle. The process was unique, but the end result appears to make him pretty similar to Greed-Ling, the (only?) difference being he doesn't have a part of Father in him, aside from Greed-Ling having two dominant souls. Bradley seemed to be on the other end of the spectrum since he seemingly only had a part of father in him, and no extra store of souls. Hence aging and no regeneration.

Fullmetal Alchemist: Brotherhood director Yasuhiro Irie has confirmed on his Twitter account on Thursday that original manga creator Hiromu Arakawa has planned out the final manga installment, and that his staff is writing and storyboarding the material for the anime based that ending. He added that he was shocked by the enormity of the final chapter and noted that there are only two months left.

I've read that the anime is ending at 62, or was it 63? either way its not to far off. Its suprising to me that the manga only has 2 chapters left, i wonder how itll be concluded after what happened last chapter. Maybe the homunculi/father win in the end, which i severely doubt.
