Nah, do all the quests. They're fun. I hate going back to the citadel everytime I move the main plot forward though. If you don't you can accidentally skip some.

I used Tali and Liara on the Hardcore playthrough I did a few weeks ago. I'll get round to Insanity eventually, just want to wait until I'm bored enough to 100% the game again so I can get another 2 of the companion bonuses. Probably Ashley and Kaidan since they have the best bonuses. Dunno whether to use my Adept again and try to get level 60. Takes like 240k XP to get to level 50 and then another 620k just to go 50-60. Managed to get from 1-51 in the one playthrough but it'd probably be quicker to just spam the main storyline quests a few times. Hopefully carrying over your save to Mass Effect 2 will do something interesting.
