    Requiring the services of someone with basic photoshop skills, FOR SCIENCE!

    Took two pictures of an actin cytoskeletion, one using phase contrast microscopy and the other using florescence microscopy and I need to overlay the the two pictures. I don't have photoshop (downloading at the very second) so any help with this will be much appreciated.

    If it's just putting each into a layer and adding transparency, I think paint.net can achieve that, but I don't have it with me right now so I can't confirmo.

    What % blending do you want, and which of the two should be the background image at 100% opacity?

    Probably 100% blending with the florescence picture as the background image. My photoshop download finished, i'll see if I can make it work, but thanks

    No matter how you layer it, overlay it, and set the opacity, you can't see the florescent because it's dull and overpowered by the picture with contrast.

    I colored the fluorescent layer. It's the only way I could make it visible.

    Tyche is right the only way to see a difference between them when you overlap is to colorize one. They need some difference in value which when they are both greyscale they do not have.

    ok a couple of things:

    1. You set too low of an exposure for both images- they are both way too dark.
    2. the phase contrast (possibly both because the flor image is so dark) is not properly in focus
    3. This is labeling of actin so ideally they should look the same. This type of staining is typically run as a control to either show all the cells or something to that effect. A positive control. What you're looking for is to show that all cells picked up the flourescent label.


    I've adjusted brightness and contrast in both (flor images typically loads of contrast, not much brightness or you get background issues). I added color to the flor image to make it stand out more and merged them. If you are putting these in a lab report or something you'd be smart to show all three so they can be compared individually and merged.

    Pretty much what Nystul said. I'm curious though... what staining label did you use for the actin?
