He's actually really well known but I didn't realize it until recently. My girl watches this dumb show called Chelsea Lately which is basically a talk show, but she has on a bunch of comedians that help her make fun of w/e they happen to be talking about. Well it turns out that John Caparulo has been part of the show basically since it started, so that's wtf happened to him.
Jo Koy is also one of my favorites

Sooo angry.
Yeah I think I've seen some clips of him from that show. That dude cracks me the fuck up. I'm trying to find the clip where he's talking about his ex or whatever just saying the meanest shit possible in an argument, but I can't find it online anywhere, they play it on Blue Collar Radio a lot though.
Old school: Eddie Murphy - Raw
New(er) School: Dave Chapelle - Killing them softly
Bill Hicks
No love for Nick Swardson?
Forgot about Mike Birbiglia too. Also hilarious
Swardson is fantastic, I forgot about him. There was this one gangly looking guy I saw on Comedy Central whom I found funny as well, but I can't remember his name. He wore a hat and had long blonde stringy hair though, I think one of his jokes was about making a resolution not to talk to stupid people anymore.
Oh and adding the prerequisite Pryor entry, though that goes without saying because he's easily #1 all-time.
Lets go asian

Most of my favorites were listed already, but I don't see the Sklar Brothers.

Stella - consists of Michael Ian Black, Michael Showalter, and David Wain, all of whom are decent individually as well
you have no idea how confused I was when I saw their comedy central special
it was like Comedy Central presents Stella
suddenly, 3 dudes.
I still don't get what's so funny about Stella. I mean, yeah it's funny in 3 straight guys acting gay, but beyond that it's really just retarded to me.
I just like the absurdity and randomness of their act. Their show (and Michael & Michael Have Issues) was a good example of what I mean, I don't find the homosexual undertones that funny.
One of the oldest and goodest of the Oldies but Goodies:
Steven Wright.
